Use the endpoint in the established procedure: repeated weighing should show that the sample's mass has stopped changing. The government method uses progressively shorter heating steps and a separate cup of water to limit burning. Follow the full procedure for your equipment rather than heating continuously until the tissue darkens. Charring changes the sample and should not be treated as evidence of a more complete water removal.
