Detached tissue can deteriorate for reasons unrelated to the pathogen, obscuring the response being tested. Mango assay development has encountered this problem with leaves and branches. Check uninoculated comparison tissue and validate how long the material remains interpretable. A collapsed sample is not automatically evidence of susceptibility, particularly if healthy detached controls decline during the same period.
